去污剂在膜蛋白研究中的应用
摘 要 膜蛋白(Membrane Proteins) 主要定位在生物膜的表面,在各种重要的生理活动中发挥着重要作用,覆盖基因调控、信号转导和能量代谢等各个层面。作为细胞中一类重要的蛋白质分子,其基因数目约占整个基因组编码基因的30%,但已知的膜蛋白结构数目却只占整个蛋白质结构数据库的1.1%左右[1]。近年来,随着新型去污剂的不断涌现以及膜蛋白表达、纯化、结晶及衍射技术的不断完善,膜蛋白结构解析日益成为结构生物学领域的研究热点。本文就去污剂在膜蛋白的分离、纯化和结晶中的应用作简要概述。

APPLICATIONS OF DETERGENTS IN MEMBRANE PROTEINS RESEARCH
Abstract Membrane proteins mainly locate on the surface of cells and organelles, which are virtually important in physical activities, including gene regulation, signal transduction and energy metabolism. Membrane proteins play significant roles in many aspects of cell biology, which account for approximately 30% of the entire genome encodes genes. However, they comprise only 1.1% of the total protein data bank (PDB) entries [1]. In recent years, within the emerging of new detergents and improving of membrane-protein related technology such as expression, purification, crystallization and diffraction, membrane proteins have become a hot spot of the structural biology. Here, we reviewed the applications of detergents in membrane protein extraction, purification, and crystallization.
